PRP Centrifuge Frequently Asked Questions

PRP centrifuge is specifically designed to stratify blood for aesthetic usage. Normally it is low speed centrifuge ( ≤4000rpm) with various angle rotors to fix centrifuge tubes. RCF & time varies with different gender and different aesthetic purpose. Low volume PRP is needed if only for anti-wrinkle, facial skin rejuvenation, face line sculpturing, pigment remove etc; large volume PRP is needed if used for FAT transplant.

The PRP separation is done in centrifuge. Different centrifuges with different specifications need to set different speed & time when prepare PRP. For our PRP centrifuge machine, the 1st centrifuge is at 3000 rpm for 6 minutes. Then we separate the erythrocytes from platelet poor plasma. The 2nd centrifuge is at 3000 rpm for 6 minutes too, then separate the PRP from platelet poor plasma.

PRP preparation need to set centrifuge speeds ranging from 2,200 RPM to 3,500 RPM. It can be used as a single or double spin system, depending on what PRP composition (type) your injection requires.Normally we recommend dual centrifuge method for PRP preparation, but only single centrifuge for PPP preparation.

The PRP centrifuge is 340mm x 280mm x 250mm (WxDxH).

The operating environment where the centrifuge is located should be 50°F (10°C) to 95°F (35°C) with a relative humidity of ≤85%.

The centrifuge should be installed on a flat, sturdy platform. Leave a large free space on the sides and back of the centrifuge for cooling and ventilation.

The KETHINK centrifuge can spin 10mL-15mL PRP tubes.

Centrifugation is to use the powerful centrifugal force generated by the high-speed rotation of the centrifuge rotor to accelerate the sedimentation speed of the particles in liquid, and separate the substances with different sedimentation coefficients and buoyancy densities in the sample.

RCF is an expression of g-force, when calculating g-force,the speed is very important when calculate this RCF which is expressed in revolutions per minute (rpm).

The relationship between the speed r/min and the centrifugal force g is not proportional, but also related to the radius. At the same speed, the radius is doubled, and the centrifugal force g is doubled. The conversion formula for the relationship between rotation speed (r/min) and centrifugal force (g value) is as follows:

G = 1.119 x (10∧- 5) * R * (rpm)2

Speaking of PRP, we must first understand growth factors first. Blood is composed of red blood cells, white blood cells, plasma and platelets. Growth factors are produced by activated cells in the hematopoietic system, immune system or inflammatory response. They are highly active and multifunctional polypeptides, proteins or glycoproteins that can regulate cell differentiation and proliferation and induce cell function.

Platelet (platelet) is a normal physiological component in human blood. When tissue is damaged, platelets quickly accumulate to vascular stumps and wounds, and release a variety of biologically active factors. These cytokines can activate the coagulation system, promote hemostasis and help wound healing. There are more than 30 kinds of cell activity factors released by platelets

PRP is a platelet concentrate extracted from whole blood by centrifugation. It is rich in high concentrations of platelets, white blood cells and fibrin.

1. PRP Advantage

  • prp is self-generating, no disease transmission, immune rejection, and concerns that heterogeneous gene products may change the human genetic structure.
  • PRP contains a variety of high-concentration growth factors, and the ratio of various growth factors is in line with the normal ratio in the body, so that various growth factors have the best synergy.
  • PRP contains a large number of white blood cells, among which mononuclear macrophages can clear local pathogens and local necrotic tissues, greatly enhancing local anti-infection ability.
  • PRP contains a large amount of fibrin, which provides a good scaffold for repairing cells and stimulates tissue regeneration.

2. Key growth factor that involved in aesthetic surgery filed

  • TGF-β
  • EGF
  • PDGF
  • VEGF
  • FGF
  • KGF
  • IGF

3. How growth factors act on target cells

  • Autocrine: Cell growth factor produces receptors for this growth factor at the same time, acting on its own cells.
  • Paracrine: A growth factor of a cell acts on another cell containing its receptor.
  • Near secretion: the growth factor directly acts on the recipient cell immediately after being released from the cell.
  • Endocrine: Growth factors act on cells at a certain distance through the pipeline

4. Applications of PRP in medical aesthetics

  • Facial skin rejuvenation: facial skin is loose, rough, dull, dry, with large pores.
  • Removal of wrinkles: Forehead lines, Sichuan word lines, crow’s feet lines, fine lines around eyes, naso dorsum lines, nasolabial lines, mouth wrinkles, neck lines.
  • Pigment diseases: improve pigmentation, pigmentation, sunburn, erythema, and chloasma.
  • Aging around the eyes: tear grooves, dark circles, fine wrinkles.
  • Anti-allergic: allergic skin, allergic skin beauty.
  • Wounds: Promote healing, fight infections, trauma and acne left by depressed scars.
  • Promote hair regeneration; treat alopecia areata, thinning hair, assist hair transplantation.

(NOTE: PRP contraindications: skin diseases (systemic lupus erythematosus, porphyria, allergies), tumors, chemotherapy, severe metabolic and mental diseases, abnormal platelet function, anticoagulant therapy, sepsis, etc. )

PRP Preparation Procedure

At present, there is no uniform standard for the specific clinical application of PRP. There are many preparation methods, and some methods are very complicated and require professional equipment.

The most common and simple method is to use a low-speed centrifuge and anticoagulation test tube. According to our long term research & comparison of the different PRP centrifuge procedure, the dual centrifugation method is recommend for our KT-TD3WS PRP centrifuge.

Step 1: draw cubital venous blood and mix well.
(Note: Try not to produce excessive air bubbles during the blood draw, so as not to damage the blood cells.)
Step 2: 1st centrifugation
Centrifuge at 3000rpm for 6 minutes.(Balance before procedure). The angle rotor is better than the horizontal rotor.
Step 3: Let stand for 10-15 minutes.

Step 4: Draw out 1/2 of the upper serum from top to bottom.
Step 5: The rest part to proceed the 2nd centrifugation. Centrifuge at 3000 rpm for 6 minutes.(Balance before procedure).
Step 6: Shake well before extraction, shake off the platelets on the separation gel and the tube wall before extracting PRP.

PRP Preparation Procedure

PRP injection therapy normally is consists of 3 treatments that lasts for 1-1.5 months. Then need a 1-1.5 months of maintenance treatments period. PRP injection therapy price varies due to the different injection area, injection period, and other factors .i.e. your country & location, facilities, and the expertise of the doctor performing the treatment. The following is a price list for the treatment.

price(USD) per treatment (for face)3 treatments (1-1.5 months)maintenance treatments((1-1.5 months)
  • Autologous, from patient’s own blood, very safe as a natural biofiller for human body;
  • The PRP therapy on face is painless, fast and safe, normally one treatment lasts for less than 1 hour;
  • Significant effect after treatment.

PRP Joint Application

  • Large-wound plastic surgery: PPR gel can be used in facial and neck lift, breast reduction, breast augmentation and other larger wounds and plastic surgery. It can reduce postoperative bleeding, hematoma, swelling and other complications, and promote the wound healing speed and the patient’s recovery is accelerated.
  •  Fat transplantation: Mixing injection of PRP with autologous fat particles purified by centrifugation can greatly improve the survival rate of autologous fat.
  • Laser: It can speed up the re-epithelialization process of the skin after laser treatment.

Microneedle Mesotherapy

Microneedle Mesotherapy is the most common PRP injection method, which can effectively reduce skin fine lines and increase skin elasticity and luster.

Operation Steps:
  • Prepare inactive liquid PRP and 5% CaCl2 0.1ml+2% lidocaine, and let it stand for 4-5 minutes;
  • Clean the face at the same time, apply ice on the surface of the treatment area for 10 minutes.
  • Disinfect the face, spread a sterile sheet, and wipe the treated area with saline.
  • Draw PRP into the syringe and perform microneedle operation while dripping liquid. If you use a mesotherapy gun, you can directly inject the liquid, depending on the treatment site to control the operation density and operation time.Usually, stop the treatment when the skin become red. The acne skin should be slightly oozing blood. Generally, the whole process can be completed in 10-20 minutes operate.
  • NOTE: Facial operation should be even and symmetrical, and the key area can increase the operation density.
PRP Filling Injection

For deeper wrinkles, PRP is also used as a filler, injects the leather shallow layer with a line / fan-shaped plane injection method.

PRP Treatment Before & After

PRP treatment is not a one-time treatment. You will find changes for each treatment. Take treatment on face before and after pictures to see difference.

PRP Treatment Aftercare

  • After the physiological saline gauze should be covered in the treatment site for 20-40 minutes, until the PRP liquid of the skin is solidified, then gently wipe it with wet yarns, and then dust and other impurities should be avoided after surgery. Do not force it hard. Injection area, 8 hours later, you can wash your face.
  • Should not taking non-steroidal anti-inflammatory drugs within 1 week before & after injection .
  • After the injection, its filling effect can be seen in 1-2 months. The effect is best in 3-6 months. The efficacy can usually last for about 1 year.

PRP Treatment Side Effects

PRP injection involves using platelets that derived from a patient’s own blood, which is why people receiving this treatment do not usually have any side effects after injection. However, it is possible to shortly swollen in the injection area after PRP filling injection. But there is no hematoma, and the length of the swelling period is determined according to the individual difference.Normal swelling time is 12-24 hours. Most people can resume their normal activities almost immediately after having a PRP injection.
